BackgroundEHD2, a member of the EH domain-containing protein family, is primarily located in the plasma membrane but can also be found in the cytoplasm and endosomes. It functions as a nuclear-cytoplasmic shuttle protein and acts as a corepressor of transcription within the nucleus to inhibit gene transcription. EHD2 plays a key role in endocytic transport, being involved in the formation and maintenance of endosomal tubules and vesicles, which are essential for intracellular transport of proteins and other substances. The N-terminus of EHD2 is attached to the cell membrane, while its C-terminus binds to actin-binding proteins, linking the actin cytoskeleton to membrane curvature and promoting cell endocytosis.
