ADAMTSL1 encodes a secreted protein resembling members of the ADAMTS (a disintegrin and metalloproteinase with thrombospondin motif) family. This protein lacks the propeptide region and the metalloproteinase and disintegrin-like domains, which are typical of the ADAMTS family, but contains other ADAMTS domains, including the thrombospondin type 1 motif. This protein may have important functions in the extracellular matrix. Alternatively spliced transcript variants have been described, but their biological validity has not been determined. The deduced 525-amino acid protein contains an N-terminal signal peptide. The mature secreted form contains 497 amino acids and has a calculated molecular mass of about 55 kD.
Human ADAMTS-like protein 1 (ADAMTSL1) ELISA Kit employs a two-site sandwich ELISA to quantitate ADAMTSL1 in samples. An antibody specific for ADAMTSL1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyADAMTSL1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ADAMTSL1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ADAMTSL1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
