Ryanodine receptors (RyRs) form a class of intracellular calcium channels in various forms of excitable animal tissue like muscles and neurons. It is the major cellular mediator of calcium-induced calcium release (CICR) in animal cells.Ryanodine receptor 2 is a ryanodine receptor found in cardiac muscle sarcoplasmic reticulum. The encoded protein is one of the components of a calcium channel, composed of a tetramer of the ryanodine receptor proteins and a tetramer of FK506 binding protein 1B proteins, that supplies calcium to cardiac muscle. Mutations in this gene are associated with catecholaminergic polymorphic ventricular tachycardia, stress-induced polymorphic ventricular tachycardia, and arrhythmogenic right ventricular dysplasia.
Human Ryanodine receptor 2 (RYR2) ELISA Kit employs a two-site sandwich ELISA to quantitate RYR2 in samples. An antibody specific for RYR2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyRYR2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for RYR2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of RYR2 bound in the initial step. The color development is stopped and the intensity of the color is measured.
