TREM2 is a membrane protein that forms a receptor signaling complex with TYROBP. The encoded protein may be involved in chronic inflammation by triggering the production of constitutive inflammatory cytokines. Defects in this gene are a cause of polycystic lipomembranous osteodysplasia with sclerosing leukoencephalopathy (PLOSL). Monocyte/macrophage- and neutrophil-mediated inflammatory responses can be stimulated through a variety of receptors, including G protein-linked 7-transmembrane receptors , Fc receptors, CD14 and Toll-like receptors , and cytokine receptors. Engagement of these receptors can also prime myeloid cells to respond to other stimuli. Myeloid cells express receptors belonging to the Ig superfamily, such as TREM2, or to the C-type lectin superfamily.
Human Triggering receptor expressed on myeloid cells 2 (TREM2) ELISA Kit employs a two-site sandwich ELISA to quantitate TREM2 in samples. An antibody specific for TREM2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTREM2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TREM2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TREM2 bound in the initial step. The color development is stopped and the intensity of the color is measured.
