Villin is a 92.5 kDa tissue-specific actin-binding protein associated with the actin core bundle of the brush border. Villin contains multiple gelsolin-like domains capped by a small (8.5 kDa) "headpiece" at the C-terminus consisting of a fast and independently-folding three-helix bundle that is stabilized by hydrophobic interactions. The headpiece domain is a commonly studied protein in molecular dynamics due to its small size and fast folding kinetics and short primary sequence.Villin is an actin binding protein expressed mainly in the brush border of the epithelium in vertebrates but sometimes it is ubiquitously expressed in protists and plants. Villin is found localized in the microvilli of the brush border of the epithelium lining of the gut and renal tubes in vertebrates.
Human Villin-1 (VIL1) ELISA Kit employs a two-site sandwich ELISA to quantitate VIL1 in samples. An antibody specific for VIL1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyVIL1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for VIL1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of VIL1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Human Villin-1 (VIL1) ELISA Kit listed herein is for research use only and is not intended for use in human or clinical diagnosis.
