Lanosterol 14-alpha demethylase (CYP51A1) has a alternative name : Cytochrome P450 Family 51 Subfamily A Member 1, This gene encodes a member of the cytochrome P450 superfamily of enzymes. The cytochrome P450 proteins are monooxygenases which catalyze many reactions involved in drug metabolism and synthesis of cholesterol, steroids and other lipids. This endoplasmic reticulum protein participates in the synthesis of cholesterol by catalyzing the removal of the 14alpha-methyl group from lanosterol. Homologous genes are found in all three eukaryotic phyla, fungi, plants, and animals, suggesting that this is one of the oldest cytochrome P450 genes. This protein acts as an enzyme. It is known to catalyze the following reaction : A 14-alpha-methylsteroid + 3 O(2) + 3 NADPH = a Delta(14)-steroid + formate + 3 NADP(+) + 4 H(2)O.
Bovine Lanosterol 14-alpha demethylase (CYP51A1) ELISA Kit employs a two-site sandwich ELISA to quantitate CYP51A1 in samples. An antibody specific for CYP51A1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyCYP51A1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CYP51A1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CYP51A1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
