The protein encoded by CD45 gene is a member of the protein tyrosine phosphatase (PTP) family. PTPs are known to be signaling molecules that regulate a variety of cellular processes including cell growth, differentiation, mitosis, and oncogenic transformation. This PTP contains an extracellular domain, a single transmembrane segment and two tandem intracytoplasmic catalytic domains, and thus is classified as a receptor type PTP. This PTP has been shown to be an essential regulator of T- and B-cell antigen receptor signaling. It functions through either direct interaction with components of the antigen receptor complexes, or by activating various Src family kinases required for the antigen receptor signaling. This PTP also suppresses JAK kinases, and thus functions as a regulator of cytokine receptor signaling.

Rat Cluster of differentiation 45 (CD45) ELISA Kit employs a two-site sandwich ELISA to quantitate PTPRC in samples. An antibody specific for PTPRC has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPTPRC present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PTPRC is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PTPRC bound in the initial step. The color development is stopped and the intensity of the color is measured.
