Type 5 acid phosphatase is a glycosylated monomeric metalloenzyme expressed in mammals. It has a molecular weight of approximately 35kDa, a basic isoelectric point (7.6-9.5) , and optimal activity in acidic conditions. TRAP is synthesized as latent proenzyme and activated by proteolytic cleavage and reduction. It is differentiated by other mammalian acid phosphatases by its resistance to inhibition by tartrate, molecular weights and characteristic purple colour. The mechanism of phosphate ester hydrolysis by TRAP is through a nucleophilic attack mechanism, whereby, catalysis occurs with the binding of a phosphate-substrate to the Fe2+ in the active site of TRAP. This is then followed by a nucleophilic attack by a hydroxide ligand on the bound phosphorus atom, resulting in cleavage of the phosphate ester bond and production of an alcohol.
Rat Tartrate-resistant acid phosphatase type 5 (ACP5) ELISA Kit employs a two-site sandwich ELISA to quantitate ACP5 in samples. An antibody specific for ACP5 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyACP5 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ACP5 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ACP5 bound in the initial step. The color development is stopped and the intensity of the color is measured.
