Neuronal acetylcholine receptor subunit alpha-7 forms a homo-oligomeric channel, displays marked permeability to calcium ions and is a major component of brain nicotinic receptors that are blocked by, and highly sensitive to, alpha-bungarotoxin. Once this receptor binds acetylcholine, it undergoes an extensive change in conformation that affects all subunits and leads to opening of an ion-conducting channel across the plasma membrane.This gene is located in a region identified as a major susceptibility locus for juvenile myoclonic epilepsy and a chromosomal location involved in the genetic transmission of schizophrenia. An evolutionarily recent partial duplication event in this region results in a hybrid containing sequence from this gene and a novel FAM7A gene.
Rabbit Neuronal acetylcholine receptor subunit alpha-7 (CHRN ELISA Kit employs a two-site sandwich ELISA to quantitate CHRNA7 in samples. An antibody specific for CHRNA7 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyCHRNA7 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CHRNA7 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CHRNA7 bound in the initial step. The color development is stopped and the intensity of the color is measured.
