Mucin 1, cell surface associated (MUC1) or polymorphic epithelial mucin (PEM) is a mucin encoded by the MUC1 gene in humans. MUC1 is a glycoprotein with extensive O-linked glycosylation of its extracellular domain. Mucins line the apical surface of epithelial cells in the lungs, stomach, intestines, eyes and several other organs. Mucins protect the body from infection by pathogen binding to oligosaccharides in the extracellular domain, preventing the pathogen from reaching the cell surface. Overexpression of MUC1 is often associated with colon, breast, ovarian, lung and pancreatic cancers. Joyce Taylor-Papadimitriou identified and characterised the antigen during her work with breast and ovarian tumors.

Pig Mucin-1 (MUC1) ELISA Kit employs a two-site sandwich ELISA to quantitate MUC1 in samples. An antibody specific for MUC1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMUC1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MUC1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MUC1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
