Ncoa5 encodes a coregulator for the alpha and beta estrogen receptors and the orphan nuclear receptor NR1D2. The nuclear receptor coactivator 5 localizes to the nucleus, and is thought to have both coactivator and corepressor functions. Its interaction with nuclear receptors is independent of the AF2 domain on the receptors, which is known to regulate interaction with other coreceptors. Two alternatively spliced transcript variants for this gene have been described. However, the full length nature of one of the variants has not been determined.

Standards and samples are pipetted into the wells and anyNCOA5 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for NCOA5 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of NCOA5 bound in the initial step. The color development is stopped and the intensity of the color is measured.
