PAPP-A is a large heterottetrameric glycoprotein of approximately 500 kDa, which was first discovered in serum from pregnant women. The hetreotetramer consists of two PAPP-A subunits and two proMBP subunits. Low maternal serum levels of PAPP-A in first trimester biochemical screening is used as a marker of Down’s syndrome. a secreted metalloproteinase which cleaves insulin-like growth factor binding proteins (IGFBPs). It is thought to be involved in local proliferative processes such as wound healing and bone remodeling. Low plasma level of this protein has been suggested as a biochemical marker for pregnancies with aneuploid fetuses (fetuses with an abnormal number of chromosomes). For example, low PAPPA may be seen in prenatal screening for Down syndrome.
Mouse Pregnancy associated plasma protein A (PAPP-A) ELISA Kit employs a two-site sandwich ELISA to quantitate PAPP-A in samples. An antibody specific for PAPP-A has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPAPP-A present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PAPP-A is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PAPP-A bound in the initial step. The color development is stopped and the intensity of the color is measured.
