14-3-3 proteins are a family of conserved regulatory molecules that are expressed in all eukaryotic cells. 14-3-3 proteins have the ability to bind a multitude of functionally diverse signaling proteins, including kinases, phosphatases, and transmembrane receptors. More than 200 signaling proteins have been reported as 14-3-3 ligands. The name 14-3-3 refers to the particular elution and migration pattern of these proteins on DEAE-cellulose chromatography and starch-gel electrophoresis. The 14-3-3 proteins eluted in the 14th fraction of bovine brain homogenate and were found on positions 3.3 of subsequent electrophoresis by Moore and Perez (1967).

Human 14-3-3 protein ELISA Kit employs a two-site sandwich ELISA to quantitate 14-3-3 protein in samples. An antibody specific for 14-3-3 protein has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any14-3-3 protein present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for 14-3-3 protein is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of 14-3-3 protein bound in the initial step. The color development is stopped and the intensity of the color is measured.
