Pancreatic polypeptide (PP), neuropeptide Y (NPY), and peptide YY (PYY) are related 36-amino acid hormones. A number of structurally related receptors for these peptides have been isolated. Bard et al. (1995) used the rat Y1 receptor (NPY1R) as a probe to clone a novel receptor, PPYR1 (termed Y4 by them), from a human genomic DNA library. PPYR1 encodes a predicted 375-amino acid polypeptide having highest homology to the Y1 receptor gene (42% amino acid identity). Using RT-PCR, they showed that the PPYR1 receptor is expressed in several human tissues, including brain, coronary artery, and ileum.Bard et al. (1995) expressed the PPYR1 gene in COS-7 cells and performed a hormone-binding assay which showed that the PPYR1 receptor binds PP, PYY, and NPY hormones.
Human Neuropeptide Y receptor type 4 (PPYR1) ELISA Kit employs a two-site sandwich ELISA to quantitate PPYR1 in samples. An antibody specific for PPYR1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPPYR1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PPYR1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PPYR1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
