Carbonyl reductase 3 catalyzes the reduction of a large number of biologically and pharmacologically active carbonyl compounds to their corresponding alcohols. The enzyme is classified as a monomeric NADPH-dependent oxidoreductase. CBR3 contains three exons spanning 11.2 kilobases and is closely linked to another carbonyl reductase gene - CBR1. Carbonyl reductase (CBR) catalyzes the reduction of a large number of biologically and pharmacologically active carbonyl compounds to their corresponding alcohols. By analyzing sequences from the 21q22.2 chromosomal region, Watanabe et al. (1998) identified genomic clones corresponding to CBR1 and to CBR3, a novel carbonyl reductase gene. The deduced 277-amino acid CBR3 protein is 84% similar to the CBR1 protein.
Human Carbonyl reductase [NADPH] 3 (CBR3) ELISA Kit employs a two-site sandwich ELISA to quantitate CBR3 in samples. An antibody specific for CBR3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyCBR3 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CBR3 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CBR3 bound in the initial step. The color development is stopped and the intensity of the color is measured.
