CYP2C8 encodes a member of the cytochrome P450 superfamily of enzymes. The cytochrome P450 proteins are monooxygenases which catalyze many reactions involved in drug metabolism and synthesis of cholesterol, steroids and other lipids. This protein localizes to the endoplasmic reticulum and its expression is induced by phenobarbital. The enzyme is known to metabolize many xenobiotics, including the anticonvulsive drug mephenytoin, benzo(a)pyrene, 7-ethyoxycoumarin, and the anti-cancer drug taxol. CYP2C8 is located within a cluster of cytochrome P450 genes on chromosome 10q24. Several transcript variants encoding a few different isoforms have been found for CYP2C8.
Human Cytochrome P450 2C8 (CYP2C8) ELISA Kit employs a two-site sandwich ELISA to quantitate CYP2C8 in samples. An antibody specific for CYP2C8 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyCYP2C8 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CYP2C8 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CYP2C8 bound in the initial step. The color development is stopped and the intensity of the color is measured.
