The type II cytokeratins consist of basic or neutral proteins which are arranged in pairs of heterotypic keratin chains coexpressed during differentiation of simple and stratified epithelial tissues. This type II cytokeratin is specifically expressed in the spinous and granular layers of the epidermis with family member KRT10 and mutations in these genes have been associated with bullous congenital ichthyosiform erythroderma. The type II cytokeratins are clustered in a region of chromosome 12q12-q13.Keratin 1 is specifically expressed in the spinous and granular layers of the epidermis with family member keratin 10. Mutations in this gene have been associated with the variants of bullous congenital ichthyosiform erythroderma in which the palms and soles of the feet are affected.
Human Keratin, type II cytoskeletal 1 (KRT1) ELISA Kit employs a two-site sandwich ELISA to quantitate KRT1 in samples. An antibody specific for KRT1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyKRT1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for KRT1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of KRT1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
