Keratin 10 (epidermolytic hyperkeratosis; keratosis palmaris et plantaris), also known as KRT10, is a human gene.This gene encodes a member of the type I (acidic) cytokeratin family, which belongs to the superfamily of intermediate filament (IF) proteins.Keratins are heteropolymeric structural proteins which form the intermediate filament. These filaments, along with actin microfilaments and microtubules, compose the cytoskeleton of epithelial cells. Mutations in this gene are associated with epidermolytic hyperkeratosis. This gene is located within a cluster of keratin family members on chromosome 17q21.
Human Keratin, type I cytoskeletal 10 (KRT10) ELISA Kit employs a two-site sandwich ELISA to quantitate KRT10 in samples. An antibody specific for KRT10 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyKRT10 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for KRT10 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of KRT10 bound in the initial step. The color development is stopped and the intensity of the color is measured.
