Laminins are basement membrane components thought to mediate the attachment, migration and organization of cells into tissues during embryonic development by interacting with other extracellular matrix components.Laminin subunit alpha-3 is the alpha-3 chain of laminin 5, which is a complex glycoprotein composed of three subunits (alpha, beta, and gamma). Laminin 5 is thought to be involved in cell adhesion, signal transduction and differentiation of keratinocytes. Mutations in this gene have been identified as the cause of Herlitz type junctional epidermolysis bullosa. Human Laminin subunit alpha-3 (LAMA3) ELISA Kit employs a two-site sandwich ELISA to quantitate LAMA3 in samples. An antibody specific for LAMA3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyLAMA3 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for LAMA3 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of LAMA3 bound in the initial step. The color development is stopped and the intensity of the color is measured.
