Galectins constitute a family of proteins that bind to β-galactoside residues and have diverse physiological functions. Report on the identification of a galectin-like molecule, galectin-12, in a human adipose tissue cDNA library. The protein contained two potential carbohydrate-recognition domains with the second carbohydrate-recognition domain being less conserved compared with other galectins.Galectin-12 mRNA was predominantly expressed in adipose tissue of human and mouse and in differentiated 3T3-L1 adipocytes. Caloric restriction and treatment of obese animals with troglitazone increased galectin-12 mRNA levels and decreased the average size of the cells in adipose tissue.
Human Galectin-12 (LGALS12) ELISA Kit employs a two-site sandwich ELISA to quantitate LGALS12 in samples. An antibody specific for LGALS12 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyLGALS12 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for LGALS12 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of LGALS12 bound in the initial step. The color development is stopped and the intensity of the color is measured.
