MED13 encodes a component of the mediator complex (also known as TRAP, SMCC, DRIP, or ARC), a transcriptional coactivator complex thought to be required for the expression of almost all genes. The mediator complex is recruited by transcriptional activators or nuclear receptors to induce gene expression, possibly by interacting with RNA polymerase II and promoting the formation of a transcriptional pre-initiation complex. The product of MED13 is proposed to form a sub-complex with MED12, cyclin C, and CDK8 that can negatively regulate transactivation by mediator.
Human Mediator of RNA polymerase II transcription subunit 13 (MED13) ELISA Kit employs a two-site sandwich ELISA to quantitate MED13 in samples. An antibody specific for MED13 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMED13 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MED13 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MED13 bound in the initial step. The color development is stopped and the intensity of the color is measured.
