Mediator of RNA polymerase II transcription subunit 13-like encoded by MED13L is a subunit of the Mediator complex, a large complex of proteins that functions as a transcriptional coactivator for most RNA polymerase II-transcribed genes. Mediator of RNA polymerase II transcription subunit 13-like is involved in early development of the heart and brain. Defects in MED13L are a cause of transposition of the great arteries, dextro-looped (DTGA).

An antibody specific for MED13L has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMED13L present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MED13L is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MED13L bound in the initial step. The color development is stopped and the intensity of the color is measured.
