MICB encodes a heavily glycosylated protein which is a ligand for the NKG2D type II receptor. Binding of the ligand activates the cytolytic response of natural killer (NK) cells, CD8 alphabeta T cells, and gammadelta T cells which express the receptor. MHC class I polypeptide-related sequence B is stress-induced and is similar to MHC class I molecules; however, it does not associate with beta-2-microglobulin or bind peptides. Alternative splicing results in multiple transcript variants.
Human MHC class I polypeptide-related sequence B (MICB) ELISA Kit employs a two-site sandwich ELISA to quantitate MICB in samples. An antibody specific for MICB has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMICB present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MICB is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MICB bound in the initial step. The color development is stopped and the intensity of the color is measured.
