MMP8 encodes a member of the matrix metalloproteinase (MMP) family of proteins. These proteins are involved in the breakdown of extracellular matrix in embryonic development, reproduction, and tissue remodeling, as well as in disease processes, such as arthritis and metastasis. Proteolysis at different sites on this protein results in multiple active forms of the enzyme with distinct N-termini. This protein functions in the degradation of type I, II and III collagens. The gene is part of a cluster of MMP genes which localize to chromosome 11q22.3. Alternative splicing results in multiple transcript variants.
Human Matrix metalloproteinase 8/Neutrophil collagenase (MMP8) ELISA Kit employs a two-site sandwich ELISA to quantitate MMP8 in samples. An antibody specific for MMP8 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMMP8 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MMP8 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MMP8 bound in the initial step. The color development is stopped and the intensity of the color is measured.
