Mucin3 encoded by MUC3 is a membrane-bound mucin that provides protection to gut epithelial cells. Mucin3 contains about 60 tandem repeats, with each repeat being around 60 aa. N-glycosylation enables the Mucin3 to localize on the cell surface, while the C-terminus interacts with the scaffold protein PDZ domain containing 1 (PDZK1). Two transcript variants, one protein-coding and the other non-protein coding, have been found for MUC3.
Human Mucin3 (MUC3) ELISA Kit employs a two-site sandwich ELISA to quantitate MUC17 in samples. An antibody specific for MUC17 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMUC17 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MUC17 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MUC17 bound in the initial step. The color development is stopped and the intensity of the color is measured.
