MYO5A is one of three myosin V heavy-chain genes, belonging to the myosin gene superfamily. Myosin V is a class of actin-based motor proteins involved in cytoplasmic vesicle transport and anchorage, spindle-pole alignment and mRNA translocation. Myosin-Va encoded by MYO5A is abundant in melanocytes and nerve cells. Mutations in this gene cause Griscelli syndrome type-1 (GS1), Griscelli syndrome type-3 (GS3) and neuroectodermal melanolysosomal disease, or Elejalde disease. Multiple alternatively spliced transcript variants encoding different isoforms have been reported, but the full-length nature of some variants has not been determined.
Human Myosin-Va (MYO5A) ELISA Kit employs a two-site sandwich ELISA to quantitate MYO5A in samples. An antibody specific for MYO5A has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMYO5A present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MYO5A is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MYO5A bound in the initial step. The color development is stopped and the intensity of the color is measured.
