MYO9B encodes a member of the myosin family of actin-based molecular motor heavy chain proteins. Myosin-IXb represents an unconventional myosin; it should not be confused with the conventional non-muscle myosin-9 (MYH9). Myosin-IXb has four IQ motifs located in the neck domain that bind calmodulin, which serves as a light chain. Myosin-IXb complex has a single-headed structure and exhibits processive movement on actin filaments toward the minus-end. Myosin-IXb also has rho-GTPase activity. Polymorphisms in MYO9B are associated with celiac disease and ulcerative colitis susceptibility. Multiple transcript variants encoding different isoforms have been found for MYO9B.
Human Myosin-IXb (MYO9B) ELISA Kit employs a two-site sandwich ELISA to quantitate MYO9B in samples. An antibody specific for MYO9B has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMYO9B present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MYO9B is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MYO9B bound in the initial step. The color development is stopped and the intensity of the color is measured.
