N4BP2 encodes NEDD4-binding protein 2 containing a polynucleotide kinase domain (PNK) near the N-terminal region, and a Small MutS Related (Smr) domain near the C-terminal region. NEDD4-binding protein 2 can bind to both B-cell leukemia/lymphoma 3 (BCL-3) and neural precursor cell expressed, developmentally downregulated 4, (Nedd4) proteins. This protein binds and hydrolyzes ATP, may function as a 5'-polynucleotide kinase, and has the capacity to be a ubiquitylation substrate. This protein may play a role in transcription-coupled DNA repair or genetic recombination. Alternative splicing results in multiple transcript variants encoding different isoforms.
Human NEDD4-binding protein 2 (N4BP2) ELISA Kit employs a two-site sandwich ELISA to quantitate N4BP2 in samples. An antibody specific for N4BP2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyN4BP2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for N4BP2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of N4BP2 bound in the initial step. The color development is stopped and the intensity of the color is measured.
