The N-acetylglutamate synthase gene encodes a mitochondrial enzyme that catalyzes the formation of N-acetylglutamate (NAG) from glutamate and acetyl coenzyme-A. NAG is a cofactor of carbamyl phosphate synthetase I (CPSI), the first enzyme of the urea cycle in mammals. NAGS may regulate ureagenesis by altering NAG availability and, thereby, CPSI activity. Deficiencies in N-acetylglutamate synthase have been associated with hyperammonemia.

Human N-acetylglutamate synthase, mitochondrial (NAGS) ELISA Kit employs a two-site sandwich ELISA to quantitate NAGS in samples. An antibody specific for NAGS has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyNAGS present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for NAGS is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of NAGS bound in the initial step. The color development is stopped and the intensity of the color is measured.
