This locus is a processed pseudogene of the transcription factor NANOG. NANOG plays a central role in regulating self-renewal in pluripotent stem cells and tumor cells. This pseudogene contains an intact open reading frame that could potentially encode a protein similar to NANOG. Although there is no evidence of transcription from this pseudogene, RT-PCR studies suggest that NANOGP8 may be expressed in some cancer cell lines. In vitro studies using a recombinant NANOGP8 protein have shown that the protein localizes to the nucleus and can promote cell proliferation, similar to NANOG.
Human Putative homeobox protein NANOGP8 (NANOGP8) ELISA Kit employs a two-site sandwich ELISA to quantitate NANOGP8 in samples. An antibody specific for NANOGP8 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyNANOGP8 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for NANOGP8 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of NANOGP8 bound in the initial step. The color development is stopped and the intensity of the color is measured.
