Nuclear receptor coactivator 1 encoded by NCOA1 acts as a transcriptional coactivator for steroid and nuclear hormone receptors. It is a member of the p160/steroid receptor coactivator (SRC) family and like other family members has histone acetyltransferase activity and contains a nuclear localization signal, as well as bHLH and PAS domains. The product of NCOA1 binds nuclear receptors directly and stimulates the transcriptional activities in a hormone-dependent fashion. Alternatively spliced transcript variants encoding different isoforms have been identified.
Human Nuclear receptor coactivator 1 (NCOA1) ELISA Kit employs a two-site sandwich ELISA to quantitate NCOA1 in samples. An antibody specific for NCOA1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyNCOA1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for NCOA1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of NCOA1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
