INTENDED USE

 
This product is used for drug susceptibility test of mycobacteria.
 

PRINCIPLE

 
The product is to test drug susceptibility of mycobacteria with ratio method. The medium contains rich nutrients and different drugs with constant concentration. In the medium, inorganic salts, carbon source, nitrogen source, and water work to stimulate the mycobacterium growth, whereas the different drugs inhibit the mycobacterium growth.
Pick mycobacteria, isolated with liquid or solid medium to adjust to a certain concentration, inoculate respectively to different drug susceptibility test L-J media and control media, incubate at 37°C for 4 week, and finally evaluate the drug susceptibility after observing the growth of mycobacteria on drug susceptibility test medium and control medium.

STORAGE AND VALIDITY

 
·     Sealed and stored at 2-8°C in a cool and dry place.
·     The validity duration is 6 months
 

SPECIMEN COLLECTION

 
·     The clinically freshly isolated colony culture (initially within 2 weeks), which does not require subculturing, is qualified for susceptibility test.
·     Over 2 weeks isolated colony and stored colony should be subcultured on L-J culture medium. Collect the grown colony after 2~3 weeks.
·     Mycobacteria obtained from isolation on logarithmic growing stage, should be diluted to 10-2 mg/ml and 10-4 mg/ml by grinding.
 

PROCEDURE

 
1. Calibration of concentration and preparation of suspension inoculation.
·     Specimen from liquid medium: centrifuge the liquid specimen at 4000g for 15 minutes, then get the sediment, which is diluted with 0.5% sterile saline, and finally obtain mycobacterial suspension with turbidity of 1mg/ml.
·     Specimen from solid medium: pick mycobacterial colonies, which are diluted with 0.5% sterile saline, then obtain mycobacterial suspension with turbidity of 1mg/ml.
·     Suspension diluted to inoculation concentration: The suspension by turbidimetry is diluted with 0.5% sterile saline.
2. Inoculation : With inoculation concentration10 -2 mg/ml and 10-4 mg/ml ready, transfer 0.01ml with inoculating loop, then inoculate respectively to drug susceptibility medium and control medium
3. Culture: Tilt medium after inoculating and keep at 37°C to culture for 24-48h with cap loose, screw back the cap to culture for another 4 week; If mycobacteria grow well in the control tube, drug susceptibility can be reported, if no mycobacteria grows, repeat the test.
 

REFERNCE VALUE


·     Susceptibility: If there are mycobacteria growing in the control tube, drug susceptibility can be reported.
·     Negative: Detection on negative control material.
