Extreme Versatility
With the Symphony Quartet™, different reaction times and protocols can be varied for each step of the synthesis allowing the instrument to be used for simultaneous production of completely independent compounds at different scales.
For example, compounds of differing lengths can be synthesized simultaneously at different scales. Synthesis on any of the channels can start or stop at any time, regardless of whether syntheses are in progress on other channels. Fast Multiplex Operation
The speed of Symphony Quartet™ allows 4 reaction vessels to produce compounds faster and in numbers comparable to other synthesizers using many more reaction vessels. When synthesizing 12-mers, for example, it is possible to produce 16 different synthetic compounds in a 48-hour period. The difference with Symphony Quartet™ is that the first four compounds are complete in 12 hours, the next four after 24 hours, and so on, rather than having to run for the entire 48 hours before any products are complete.
The core of the Symphony Quartet™ is an innovative, highly reliable patented multiplexing matrix valve system that permits reagents to be transferred to different reaction vessels (RVs) without cross-contamination. For example, as Solvent 1 is transferred to RV1, the matrix valve block can be directing Solvent 2 to RV2, any one of 20 reagents to RV3, and nitrogen to RV4 for mixing; all simultaneously, without interference. Couplings in the reaction vessels proceed independent of each other; each according to its own unique program and protocol.
Low Coupling Cost
Amino acids for synthesis are dispensed from common bottles specific to each Amino acid. Bulk reagents can be used to prepare solutions for the 20 amino acid bottles, contributing to a low reagent cost. The fact that reaction vessels are agitated during coupling enables the synthesis of longer compounds and allows the solution to be only moderately concentrated, permitting easy dissolution of reagents.
Variable Scales
The Symphony Quartet™ offers a variable range of synthesis scales from 0.005 mmol to 0.300 mmol per channel. This allows for the economical synthesis of either small (mg) or large (near gram) quantities of compounds.
Automated Cleavage
Automated cleavage delivers the product compound from each synthesis directly to a centrifuge tube in a vented compartment, ready for further work-up and purification.
Easily Customized Protocols
Control software runs on an IBM-compatible PC and allows sequences to be created directly or imported from other programs. Partial sequences can be cut and pasted to appropriate channels, then edited as desired. Each channel is controlled independently, and custom coupling protocols can be programmed channel-by channel and residue-by-residue.

Protein Technologies, Inc. (PTI) was founded in 1985 by researchers affiliated with the University of Arizona in Tucson. In 1990, we launched our first peptide synthesizer, the PS3™, and since then we have manufactured and sold the world's finest solid phase synthesizers.
The PS3™ peptide synthesizer met with immediate success and continues to be one of the most reliable, cost effective, and easy to use instruments on the market today.
In 1993 we introduced the Symphony®, a fully automated peptide synthesizer featuring 12 independent reaction vessels that has dominated the multi-channel solid-phase synthesis market. The Symphony® is ideal for custom peptide synthesis because individual reaction vessels may be started, paused, or stopped regardless of what the other reaction vessels are doing, making it equivalent to 12 separate synthesizers in one! The Symphony® is widely regarded as perhaps the finest synthesizer available, and there are hundreds of Symphonys®operating worldwide.
In 1996 we introduced the Sonata®, a large scale automated peptide synthesizer. We included the well-established fluid movement of the Symphony®, as well as its ease of use, reagent optimization, and automated cleavage. We also added a mechanical agitator to assist nitrogen bubbling in order to efficiently mix large amounts of resin in the larger reaction vessels. The Sonata® can produce from grams to near-kilograms of expected product. In 2009, we introduced the Sonata® XT which doubles the capacity of the original Sonata® peptide synthesizer!
In 2000 we introduced the Quartet™, a 4 channel version of the Symphony® peptide synthesizer. It has all of the advantages of the Symphony® and it can be upgraded to the full Symphony® if the user's needs should grow.
In 2006, we introduced the Prelude®, a 6-channel parallel peptide synthesizer that features no-prime deliveries and extra amino acid bottles for expensive monomer additions. It can also rinse your reaction when an error occurs and send an E-mail status report to your computer or cell phone.
In 2007, we introduced the Tribute®, a revolutionary 2-channel parallel peptide synthesizer featuring a full-color chemical-resistant touchscreen, a 101-position amino acid vial rack, amino acid barcode scanning, fully-adjustable vortex and nitrogen mixing, as well as individual user logging. It also features E-mail notification and washing on error like the Prelude®. In 2009, we added the unique IntelliSynth™ UV-Monitoring and Feedback Control System to the Tribute® peptide synthesizer, which is the only UV-Monitoring system that monitors during the deprotection reaction, so it can control both the deprotection time as well as the number of repeats. In 2012, we added infrared (IR) heating to the Tribute®, giving it the fastest time to temperature of any heating technology on the market today! In addition, unlike other rapid heating methods, infrared heating can be performed on more than one reaction vessel at once, and is compatible with vortex mixing, so you can be sure you have an even temperature profile throughout your reaction vessel. Other heated systems that rely on nitrogen mixing may have hot and cold spots, which could compromise the consistency of your chemistry.
In 2010, we introduced the Overture™ robotic peptide library synthesizer. The Overture™ is the most flexible robotic platform on the market today. In addition to powerful library generation tools, the Overture™ is the only robotic platform that supports multiple, simultaneous protocols, the synthesis of different scales at the same time, and on-board automated cleavage without user intervention!
In 2012, we introduced the Symphony® X. With twice the number of reaction vessels as the original Symphony®, the Symphony® X can synthesize up to 12 peptides using preactivation or up to 24 peptides using in situ activation! It also features up to 40 amino acid positions, no-prime deliveries for expensive reagent additions, infrared heating, UV-monitoring, E-mail notification and more! With its 12 independent reactor pairs, the Symphony® X is the perfect instrument for peptide production or protocol optimization.
